COMMON ERRORS IN PHARMACEUTICAL PURIFIED WATER SYSTEMS

1. Fluctuations in feed water quality

The feed water source in Vietnam is becoming increasingly polluted, and the characteristics of the feed water change over time. The purified water production line must be capable of handling fluctuations in inlet water quality.

2. High investment and operating costs

“Over-engineering” should be avoided, as it makes the system more expensive and harder to maintain and service.

  • Sometimes systems are designed much more complexly than necessary for the purpose of TOC control (including treated water TOC and CIP line TOC).
  • Compliance with EU GMP Annex 11 and FDA 21 CFR Part 11 is sometimes confused with the requirement that SCADA is mandatory for all systems.

3. Improper design causing secondary contamination

Improper design of purified water storage tanks will lead to an increase in the TOC index in the water: usually due to inappropriate material selection and ventilation design.

STAGE 1: PRE-TREATMENT – THE FOUNDATION TO PROTECT THE ENTIRE SYSTEM

This is an important step to treat coarse impurities and chemicals in the source water, protecting the “heart” of the system, the RO membrane, from clogging and damage.

  • Multi-media filtration: Removes suspended solids, sediment, and turbidity.
  • Activated carbon: Thoroughly adsorbs free Chlorine – an oxidizing agent that destroys the RO membrane structure. It also removes color, odor, and organic compounds.
  • Water softening: Removes Calcium (Ca²⁺) and Magnesium (Mg²⁺) ions – the main causes of scaling on the RO membrane surface, helping the system operate stably and extending its lifespan.

STAGE 2: FINE TREATMENT – THE HEART OF THE SYSTEM

  • RO Reverse Osmosis System (Single or Double Pass): The pre-treated water will be pumped at high pressure through microscopic RO membranes. This technology can remove up to 99.9% of dissolved ions, bacteria, and viruses, producing high-purity water that meets most PW requirements. We prioritize using a Double Pass RO system to ensure the highest stability in output water quality.

STAGE 3: DEEP DEMINERALIZATION – ACHIEVING ABSOLUTE PURITY

Although the RO-treated water is very clean, to achieve the extremely low electrical conductivity required by GMP-EU standards (< 1.3 µS/cm), a final deionization step is necessary.

  • EDI (Electrodeionization) Technology: This is the most advanced and popular technology today. EDI uses an electrical current to continuously remove remaining ions, helping the water achieve a very high level of purity.

Advantages: Automatic and continuous operation, highly stable water quality, and absolutely no chemical regeneration (acid/caustic) required, making it safe for production and environmentally friendly.

  • (Alternative option) Mixed-bed Ion Exchange: A traditional technology that can still be used to optimize initial investment costs, but requires stopping the system for chemical regeneration.

STAGE 4: STERILIZATION ASSURANCE AND DISTRIBUTION

  • UV Sterilization: The treated water is irradiated with UV light to destroy the DNA structure of any remaining microorganisms, preventing their growth.
  • Microfiltration (0.2 µm): This is the final safety “checkpoint” before entering the storage tank, completely removing bacterial carcasses and the smallest microparticles.
  • Storage and Recirculation Distribution: Purified water is stored in 316L sanitary stainless steel tanks and continuously circulated in a closed loop to points of use. This process ensures consistent water quality at all usage points and prevents re-contamination.

ADVANCED STEP (Optional): DISTILLATION FOR WATER FOR INJECTION (WFI)

  • For production lines of injectable drugs and vaccines, purified water (PW) will be passed through a Multi-Effect Distillation System to thoroughly eliminate bacterial endotoxins (pyrogens/endotoxins), producing WFI that meets the highest Pharmacopoeia standards.
